The PrEST antigens are used for immunization. The resulting polyclonal antibodies are affinity purified against the corresponding PrEST Antigens in a three-step procedure to remove the tag-specific antibodies and to catch the PrEST-specific.
PrEST Antigen Design
The 50-150 protein specific fragments are selected using a proprietary software to contain unique epitopes present in the native protein suitable for triggering the generation of antibodies of high specificity.
The high specificity is achieved by a complete human genome scanning to ensure that regions with the lowest homology to other human proteins are used as antigens for the generation of antibodies.
PrEST Antigen Production
For cloning of the PrESTs, a pool of RNA consisting of material from a number of human tissues is used in an RT-PCR approach. The amplified gene fragments are cloned and sequence verified. An E. coli recombinant protein expression system is used for expressing the clones and the products are purified using nickel-containing matrices (IMAC).
PrEST Antigen Validation and Quality Control
The expressed PrEST Antigens are validated using ESI mass spectrometry. Purity is analyzed using SDS page and the PrEST Antigen amount is being quantified using the Nanodrop system.

Each Triple A Polyclonal antibody has been used for IHC-staining of samples from 708 individuals originating from normal and cancer afflicted human tissues. Trained pathologists have evaluated the reliability of all IHC-staining patterns. In addition ICC-IF-based confocal microscopy has been used to study subcellular distribution patterns in selected cell-lines. Further, Triple A Polyclonals are characterized by Western Blot (WB), primarily in endogenous protein lysates from human tissues.
Triple A Polyclonals combine optimal specificity with superior versatility across applications including IHC, IF and WB. The name PrecisA Monoclonals originates from our core values at Atlas Antibodies and it also reflects our Swedish origin. The Swedish word “precisa” means precise, accurate and targeted. We find this a perfect description for our in-house developed mouse monoclonal antibodies.

The characterization of PrecisA Monoclonals starts with an extensive literature search to select the most accurate, relevant and clinically significant tissues to use for immunohistochemistry (IHC) characterization. We often present several tissue types in the IHC application data in our product catalog. In addition to positive stained tissue we also display a negative control tissue staining and if relevant clinical cancer tissue staining
The characterization for Western blot (WB) data follows that same working procedure and starts with a profound literature search to find the best matching lysate.
